Talk to Deputy Willem outside the Abbey first, to get a small quest to talk to Milly.

Collect 8 crates of grapes from the vineyards, across the river to the East.

This is likely to be the characters first encounter with mobs which will attack on sight (red target ring rather than yellow).
In order to collect the grapes, you have to right-click on the baskets out in the vineyard fields. They will then open, allowing you to loot the grapes. Note that it takes a few seconds for them to open, so you have to clear the area of any Defias first. The baskets are all over the vineyard area and respawn very fast in various spots.
Pick up the 
[4] Brotherhood of Thieves quest before heading over to the vineyards, as both quests are set in the same location.
